基于网络药理学及分子对接探究通脉降糖胶囊治疗糖尿病周围神经病变作用机制
Exploration on the Mechanism of Tongluo Jiangtang Capsules in Treating Diabetic Peripheral Neuropathy with Based on Network Pharmacology and Molecular Docking

Abstract
Objective To explore the mechanism of Tongluo Jiangtang Capsules in treating diabetes peripheral neuropathy(DPN)through network pharmacology and molecular docking.Methods Targets related to DPN were retrieved from GeneCards,DisGeNET,OMIM databases,and TCMSP and HERB databases were used to obtain the active components and action targets in Tongluo Jiangtang Capsules.The intersection targets of disease and drug targets were obtained using the Venny 2.1.0.STRING database was used to construct protein-protein interaction network,and then GO and KEGG enrichment analysis of intersection targets was performed using DAVID database.The Chinese materia medica-active components-intersection targets network diagram was drawn in Cytoscape 3.8.0 software.The core targets and core active components were screened in descending order of degree value.Then the key targets and core active components were imported into AutoDock 1.5.7 software for molecular docking verification.Results A total of 2 043 disease targets with DPN were obtained,209 active components and 466 action targets were screened,and 194 intersection targets were identified.The top 3 targets were TNF,AKT1 and IL6,mainly involved in the regulation of PI3K-Akt,AGE-RAGE,MAPK and other pathways.The molecular docking results showed that TNF bound well to luteolin,quercetin,kaempferol,and crocetin,IL6 bound well with quercetin,luteolin and kaempferol.Conclusion Tongluo Jiangtang Capsules can treat DPN by targeting core targets such as TNF,AKT1 and IL6 and activating multiple signaling pathways including PI3K-Akt,AGE-RAGE and MAPK.关键词
