化瘀解毒方通过JAK/STAT通路抑制肺癌增殖的机制研究
Mechanism of Huayu Jiedu Formula in Inhibiting Lung Cancer Proliferation via the JAK/STAT Pathway

Abstract
Objective:To investigate the mechanism by which the Huayu Jiedu Formula inhibits lung cancer proliferation based on the JAK/STAT signaling pathway.Methods:Thirty-two C57BL/6J mice were randomly divided into four groups:solvent control group,high-dose Huayu Jiedu Formula group,medium-dose Huayu Jiedu Formula group,and cyclophosphamide group,with 8 mice in each group.A lung cancer mouse model was established by subcutaneously injecting 0.5 mL(1×107 cells/mL)of Lewis cell suspension into the right forelimb axilla.The medium-and high-dose Huayu Jiedu Formula groups were administered oral gavage at 15 g·kg-1·d-1 and 30 g·kg-1·d-1,respectively,once daily.The cyclophosphamide group received intraperitoneal injections of 5 mg/mL cyclophosphamide solution twice weekly.All groups were treated for 8 consecutive days.Body weight was measured three times a week.On the 8th day,body weight,tumor volume,and tumor weight were measured to calculate the relative tumor volume(RIV),relative tumor proliferation rate(TIC),and tumor inhibition rate(IR).Real-time quantitative PCR(RT-PCR)was used to detect mRNA expression of Janus kinase 2(JAK2),signal transducer and activator of transcription 3(STAT3),STAT5,and apoptosis regulatory factor(Bax)in tumor tissues.Immunohistochemistry(IHC)and Western blot were employed to detect protein levels of JAK2,P-JAK2,STAT3,P-STAT3,STAT5,P-STAT5,and Bax in tumor tissues.Results:Observation of tumor-bearing mice revealed that compared to the solvent control group,the Huayu Jiedu Formula had no significant effect on body weight,while the cyclophosphamide group significantly reduced body weight(P<0.05),indicating the lower toxicity of the Huayu Jiedu Formula.Tumor volume and tumor weight were significantly reduced in the Huayu Jiedu Formula groups(P<0.05).RT-qPCR results showed that the Huayu Jiedu Formula did not affect JAK2,STAT3,STAT5,or Bax mRNA levels.IHC and Western blot results indicated that the Huayu Jiedu Formula inhibited the expression of P-JAK2,P-STAT3,and P-STAT5(P<0.05,P<0.01)and promoted Bax protein expression(P<0.05,P<0.01),thereby promoting apoptosis of lung cancer cells.Conclusion:The Huayu Jiedu Formula may inhibit lung cancer cell proliferation by downregulating phosphorylation levels of JAK-STAT signaling pathway-related proteins.关键词
