逍遥散加味颗粒对肝纤维化大鼠ET-1、iNOS的影响
Effects of Modified Xiaoyao Granules on ET-1 and iNOS in Rats with Liver Fibrosis

Abstract
Objective:To observe the effects of Modified Xiaoyao Granules on serum levels of endothelin-1(ET-1)and inducible nitric oxide synthase(iNOS)in rats with liver fibrosis,and to explore its molecular mechanism for treating liver fibrosis.Methods:A total of 78 Sprague-Dawley(SD)rats,with equal numbers of males and females,were randomly divided into a normal group(12 rats)and a model group(66 rats).Liver fibrosis was induced in the model group via subcutaneous injection of carbon tetrachloride,after which the rats were divided into a model control group(12 rats),high-dose(13 rats)and low-dose Modified Xiaoyao Granules(13 rats)groups,Fufang Biejia Soft Liver Tablets group(13 rats),and Fuzheng Huayu Capsule group(12 rats).The model control group received regular feed and water without treatment.The other groups were administered corresponding doses of each drug by gavage.The high-dose Modified Xiaoyao Granules group received 16 g/kg/day,while the low-dose group received 8 g/kg/day.The Fufang Biejia Soft Liver Tablets and Fuzheng Huayu Capsule groups received 0.5 g/kg/day of their respective medications.Each group was dosed once daily for four weeks.After fasting for 8 hours following the final dose,blood samples were collected from the inner canthus of the rats,and liver tissue samples were collected after fasting for 24 hours,under anesthesia.Liver tissue changes were observed with hematoxylin and eosin(HE)staining,and serum ET-1 and iNOS levels were measured using enzyme-linked immunosorbent assay(ELISA).Results:Compared to the normal group,the model control group exhibited a significant increase in ET-1 expression(P<0.05).Both high-dose and low-dose Modified Xiaoyao Granules groups showed reduced ET-1 expression compared to the model control group(P<0.05,P<0.01),with the high-dose group showing the most pronounced effect(P<0.01).There was no statistically significant difference in iNOS expression between the model control group and the high-dose Modified Xiaoyao Granules group compared to the normal group(P>0.05).Compared to the model control group,iNOS levels were reduced in the low-dose Modified Xiaoyao Granules group,the Fuzheng Huayu Capsule group,and the Fufang Biejia Soft Liver Tablets group(P<0.05,P<0.01),with the Fufang Biejia Soft Liver Tablets group showing the most notable effect.Conclusion:Modified Xiaoyao Granules show a significant anti-fibrotic effect on the liver,likely involving ET-1 and iNOS in reversing liver fibrosis.关键词
