摘要

Abstract

Objective:To investigate the reparative effect of raspberry proanthocyanidins on reproductive damage in male diabetic silkworms.Methods:Silkworms in the experimental group were fed a high-glucose diet containing 10%glucose for 18 hours to establish a type 2 diabetes model.The diabetic silkworms were randomly divided into a model group,and high-dose(50 μL),medium-dose(25 μL),and low-dose raspberry proanthocyanidins(12.5 μL)groups.The levels of testosterone(T),luteinizing hormone(LH),and follicle-stimulating hormone(FSH)in the hemolymph were measured;triglyceride(TG)levels in the fat body were determined;superoxide dismutase(SOD),malondialdehyde(MDA),and glutathione peroxidase(GSH-Px)levels in the testicular tissue were measured;pathological damage to testicular tissue was observed via HE staining;and the expression levels of siwi1 and siwi2 genes in the PiRNA signaling pathway were analyzed using RT-PCR.Results:Compared to the model group,all raspberry proanthocyanidin doses led to a reduction in blood glucose levels(P<0.05)and an increase in LH levels(P<0.05),with high-and medium-dose groups also showing increased T levels(P<0.05).The levels of GSH-Px and SOD were elevated(P<0.05),while MDA levels were decreased(P<0.05)in all raspberry proanthocyanidin groups.Testicular pathological damage was alleviated,and mRNA levels of siwi1 and siwi2 were increased(P<0.05)in all raspberry proanthocyanidin groups.Conclusion:Raspberry proanthocyanidins can improve male reproductive damage caused by diabetes by lowering blood glucose and lipid levels,increasing LH and T levels,reducing oxidative stress in the testes,alleviating testicular pathological damage,and enhancing the expression of related genes in the PiRNA signaling pathway.
