基于cAMP-Epac信号通路探讨喘咳平颗粒对哮喘幼龄大鼠气道炎症与气道重塑的作用
Effects of Chuankeping Keli(喘咳平颗粒)on airway inflammation and remodeling in juvenile asthmatic rats based on the cAMP-Epac signaling pathway

Abstract
Objective To observe the mechanism of Chuankeping Keli(喘咳平颗粒)in reducing airway inflammation and improving airway remodeling in juvenile asthmatic rats.Methods Twenty SD rats aged 6-8 weeks were randomly divided into a control group,model group,Western medicine group,and Chuankeping Keli group.An ovalbumin(OVA)-induced asthma model was established(except for the control group).After successful modeling,Chuankeping Keli and dexamethasone were administered by gavage,while the control and model groups received equal volumes of saline.Lung tissues were subjected to hematoxylin-eosin(HE)staining to assess tissue damage,Masson staining to evaluate subepithelial collagen deposition,and immuno-histochemistry to detect α-smooth muscle actin(α-SMA)levels.Enzyme-linked immunosorbent assay(ELISA)was used to measure interleukin(IL)-1β,IL-4,IL-8,and tumor necrosis factor-α(TNF-α)levels in bron-choalveolar lavage fluid(BALF).Western blot(WB)was performed to detect cyclic adenosine monophosphate(CAMP)and camp-activated exchange protein(Epac)expression.Results After Chuankeping Keli interven-tion,lung tissue damage in asthmatic rats decreased,while the wall thickness(Wd)/perimeter of basement membrane(pbm),smooth muscle area(WAm)/pbm,and smooth muscle thickness(Md)/pbm significantly in-creased(P<0.05).Compared with the model group,the Chuankeping Keli and Western medicine groups showed lower collagen and α-SMA levels in lung tissue,reduced IL-1β,IL-4,IL-8,and TNF-α levels in balf,and higher cAMP,Epac1,and Epac2 expression(P<0.05).Compared with the Western medicine group,the Chuankeping Keli group exhibited significantly lower Wd/pbm(P<0.05),decreased IL-1β,IL-4,IL-8,and TNF-α levels(P<0.05),and higher Epac1 and Epac2 expression(P<0.05).Although collagen deposition de-creased and α-SMA levels increased,the differences were not statistically significant(P>0.05).Conclusion Chuankeping Keli can alleviate airway inflammation and remodeling in juvenile asthmatic rats by activating the cAMP-Epac pathway.关键词
