Correlation between the expression of CD93 and tumor immuity in pancreatic cancer and its prognostic significance

Abstract
Objective To investigate the expression of CD93 in pancreatic cancer,its correlation with the infiltration level of immunocytes in tumor tissues,and its influence on patients'prognosis.Methods The GEPIA2 database was used to analyze the expression of CD93 in pancreatic cancer,while the TIMER database was employed to examine the correlation between CD93 expression and tumor-infiltrating immune cells.Single-cell sequencing data from the TISCH database were analyzed to explore the expression and distribution of CD93 in the tumor microenvironment of pancreatic cancer.The clinical data,tumor tissue and paired adjacent normal tissue samples were collected from 90 patients with pancreatic ductal adenocarcinoma(PDAC)who underwent surgical treatment at the Second Affiliated Hospital of Jiaxing University from Jan.2012 to Dec.2021.The immunohistochemical(IHC)staining was performed,and the immunoreactive score(IRS)system was applied to evaluate the CD93 expression level.Based on the IRS,patients were divided into the high-expression group(IRS≥3,n=41)and the low-expression group(IRS<3,n=49).The t-test was used to analyze the difference in CD93 expression level between pancreatic cancer tissues and adjacent normal tissues.Kaplan-Meier survival analysis was conducted to compare the difference in postoperative overall survival between the two groups.The univariate and multivariate Cox regression model were conducted to assess the prognostic factors of the patients.Results The GEPIA2 database analysis showed a higher level of CD93 in tumor tissues than that in paired adjacent normal tissues.The TIMER database analysis revealed a positive correlation between the CD93 expression level and the infiltration level of B cells,CD8+T cells,CD4+T cells,macrophages,neutrophils and dendritic cells in pancreatic cancer tissues(correlation coefficient>0.3,P<0.05).The single-cell sequencing analysis from the TISCH database indicated that CD93 was primarily expressed in endothelial cells and monocyte-derived macrophages within the pancreatic cancer tumor microenvironment.Among the 90 PDAC patients,the CD93 expression level was significantly higher in tumor tissues compared to adjacent normal tissues[IRS:(2.844±2.298)vs(1.822±1.466),P<0.001].Survival analysis demonstrated that the low-expression group had a longer postoperative overall survival than the high-expression group(P=0.014),and the median survival was 29 months compared to 15 months(P=0.001).Multivariate Cox regression analysis confirmed that a higher CD93 expression level was an independent risk factor of pancreatic cancer patients.Conclusion CD93 expression is positively correlated with the infiltration level of immunocytes in the pancreatic cancer.CD93 is an independent risk factor for prognosis in patients with pancreatic cancer.关键词
