急性胰腺炎小鼠模型NETs的形成和调节及其与IP6K1的相关性
The formation and regulation of NETs in a mouse model of acute pancreatitis and its correlation with IP6K1

Abstract
Objective To analyze the correlation between inositol hexaphosphate kinases 1(IP6K1)and neutrophil extracellular traps(NETs)in a mouse model of acute pancreatitis(AP).Methods Twenty wild-type C57 mice were randomly divided into Group A(non-IP6K1 defects,AP modeling,n=10),Group D(non-IP6K1 defects,non-AP modeling,n=10);Twenty C57BL/6 mice with IP6K1 gene defects were randomly divided into Group B(IP6K1 defect,AP modeling,n=10)and Group C(IP6K1 defect,non-AP modeling,n=10).HE staining was used to detect the pathological changes among 4 groups,fully automated biochemical analyzer and immunofluorescence experiments were used to analyze the myeloperoxidase(MPO),chemokine 1,NETs,and histone 3 levels in the pancreas,as well as amylase,MPO,NETs,histone 3 levels in peripheral blood.Results The pathological results of mouse pancreatic tissue showed that,severe edema and necrosis occurred in Group A,with glandular structure damage and a large amount of inflammatory cell infiltration.The edema and necrosis in Group B were milder than those in Group A,with a small amount of inflammatory cell invasion.Group C and D had normal structure.The results of the fully automated biochemical analyzer and immunofluorescence experiment showed that,a large number of NETs were formed in Group A,and the levels of MPO,chemokine 1,NETs,and histone 3 in the pancreas were higher than those in group B(P<0.05);Group B had minimal formation of NETs,and the expression levels of MPO,chemokine 1,NETs,and histone 3 in the pancreas were higher than those in Group C or Group D(P<0.05);NETs did not form in group C or group D.The levels of amylase,MPO,NETs,and histone 3 in the peripheral blood of Group A were higher than those in Group B(P<0.05),and the level of NETs in the pancreas was higher than that in peripheral blood(P<0.05).The levels of amylase,MPO,NETs,and histone 3 in peripheral blood of Group B were higher than those of group C or Group D(P<0.05).Conclusion This experimental study shows that,IP6K1 can promote the formation of NETs in AP mice,and regulate the level of NETs in pancreas and peripheral blood.This result may be one of the mechanisms of pancreatic tissue injury.关键词
