Abstract

Objective To prepare curcumin(CUR)therapeutic deep eutectic solvent nanostructured lipid carriers(CUR-NLCs)and investigate their antioxidant properties and zebrafish caudal fin repair activities.Methods Curcumin therapeutic deep eutectic solvent(DES-CUR)was prepared by heating method that utilized lysine and glycerol as raw materials and solubilizing carriers for CUR.The solubility enhancement of CUR via deep eutectic solvent systems was assessed through in vitro solubility experiments.The CUR-NLCs were then developed using nanoemulsion template technology,and their formulation was optimized based on particle size,zeta potential,and polydispersity index(PDI).The encapsulation rate,infrared and thermogravimetric analyses,and in vitro drug release behavior of the CUR-NLCs were evaluated.Finally,the antioxidant activity of CUR-NLCs was determined by DPPH and ABTS in vitro antioxidant assays;and a model of traumatic inflammation in zebrafish with cut tail was used to evaluate the efficacy of CUR-NLCs on zebrafish caudal fin repair.Results The saturation solubility of DES-CUR in ionic water was about 527 times that of free CUR.The obtained CUR-NLCs had an average particle size of 155.8 nm,a PDI of 0.276,a Zeta potential of-27.7 mV,and an encapsulation efficiency of 90.8%.The nanostructured lipid carriers showed good stability at 4℃within 28 days.At the same CUR concentration,CUR-NLCs showed slightly higher scavenging of DPPH radicals,which was 1.02 times that of free CUR;the scavenging rate of ABTS radicals by CUR-NLCs reached 98.98%,which was much higher than that of free CUR(64.45%).The concentration of CUR-NLCs at 0.75 μmol/L had the greatest effect on zebrafish caudal fin repair,and the caudal fin raw length was 51.43 μm.Conclusion DES-CUR prepared by heating method can effectively enhance the solubility of CUR,and DES can be regarded as a new strategy for effective delivery of APIs.The nanostructured lipid carriers can improve the stability and bioavailability of CUR and provide reference value for the development of new dosage forms of CUR.
