Abstract

Objective To optimize the preparation process of Domperidone nanosuspension stabilized with glycyrrhizic acid and evaluate its in vitro and in vivo properties.Methods Using Domperidone as a model drug,Domperidone nanosuspension was prepared by the anti-solvent precipitation method;the formulation process was optimized through orthogonal experiments,followed by physicochemical characterization of the resultant nanosuspension;the in vitro release of Domperidone nanosuspension was measured using the dialysis bag method;freeze-drying method was used for solidification,and particle size and polydispersity index were used as evaluation indicators to screen freeze-drying protectants;the gastrointestinal motility experiment of mice was carried out to investigate the effect of Domperidone nanosuspension on gastrointestinal motility.Results The particle size of the Domperidone nanosuspension was approximately 160 nm,with a spherical morphology,and it demonstrated good stability over a period of 30 days.The Domperidone in the nanosuspension exhibited an amorphous state,with a solubility of 59.6 µg/mL.It demonstrated a high cumulative release rate of 93.92%in simulated gastric fluid over 24 h and significantly enhanced gastrointestinal motility in mice.Conclusion Domperidone nanosuspension can effectively improve the solubility of Domperidone.
