夫西地酸脂肪酸囊泡的透皮吸收及抗痤疮疗效的初步研究
Preliminary investigation of the transdermal penetration and anti-acne efficacy of budesonide fatty acid vesicles

Abstract
Objective To investigate the in vitro permeability of fatty acid vesicles of fusidic acid(FA-FAV)and their efficacy in an acne model mouse.Methods The characterization of FA-FAV was performed using appearance features,transmission electron microscopy,particle size distribution,zeta potential,encapsulation efficiency,drug loading,differential scanning calorimetry(DSC),and Fourier transform infrared spectroscopy(FTIR).In vitro transdermal investigations were performed by utilizing Franz diffusion cells,and the skin irritation potential of FA-FAV was assessed through dermatological safety experiments.An acne model was established in mice,and changes in ear lesion morphology were observed visually,with hematoxylin-eosin(HE)staining used for pathological analysis of tissue morphological changes.Results The average particle size of the prepared FA-FAV was(146.20±0.87)nm,with a PDI of 0.132±0.002 and a Zeta potential of(-32.80±0.11)mV.The appearance and electron microscopy images showed that FA-FAV was a light blue,clear,and transparent homogeneous liquid,exhibiting a nearly spherical shape with uniform size and a relatively even dispersion.The DSC and FTIR analyses verified the effective incorporation of the medication into the vesicles.In vitro penetration and retention experiments demonstrated that FA-FAV effectively enhanced the transdermal absorption capacity of FA,with the transdermal release curve fitting the Higuchi equation.The skin safety evaluation experiment confirmed that FA-FAV was non-irritating to the skin and had good safety.The pharmacological studies indicated that the FA-FAV group significantly improved the inflammatory condition in acne-prone mice compared to the model group;the result of HE staining revealed a notable reduction in epidermal thickness of the mouse ear tissue,accompanied by less inflammatory cell infiltration.Conclusion Fatty acid vesicles can increase the permeability of FA for topical application on the skin,reducing the skin's barrier to the drug and achieving anti-acne effects,thus providing a new strategy for the clinical application of acne treatment.关键词
