Ursolic acid induces apoptosis of human colorectal cancer HT-29 cells by antagonizing Bcl-2
摘要
Abstract
Objective To investigate the effect of ursolic acid(UA)on the apoptosis of human colon cancer HT-29 cells and apoptosis-associated protein B-lymphoblastoma 2(Bcl-2).Methods HT-29 cells were cultured in vitro with UA intervention,cell viability was detected by MTT method,apoptosis was detected by flow cytometry,gel electrophoresis and DAPI staining,mitochondrial membrane potential(JC-1 staining)and cytochrome C release were determined by flow cytometry,and Bcl-2 expression was determined by Western blot.The activities of cysteinyl aspartate-specific proteinase-3(Caspase-3)and Caspase-9 were detected by colorimetry.The interaction between Bcl-2 and UA was detected by pore-forming activity technique and surface plasmon resonance technique.Bcl-2 overexpressing HT-29 cells were constructed.After the intervention of UA,the expression of Bcl-2 was determined by RT-PCR and Western blot,and the apoptosis rate was detected by DAPI staining and flow cytometry.Results UA induced apoptosis of HT-29 cells,led to loss of mitochondrial membrane potential and increased release of cytochrome C,inhibited the expression of Bcl-2,and increased the activities of Caspase-3 and Caspase-9(P<0.05).UA could enhance the permeability of cell membrane and bind specifically to Bcl-2.UA inhibited the expression of Bcl-2 in Bcl-2-overexpressed HT-29 cells and increased the apoptosis rate(P<0.05).Conclusion UA can induce apoptosis of HT-29 cells by antagonizing Bcl-2.关键词
