非编码RNA与广州人群2型糖尿病患病风险的病例对照研究
Case-control study of non-coding RNA and type 2 diabetes risk in the Guangzhou population

Abstract
Objective To explore the impact of expression levels of microRNA(miRNA),long noncoding RNA(lncRNA),and circular RNA(circRNA)on the incidence of type 2 diabetes mellitus(T2DM)in the Guangzhou population.Methods The study population consisted of community health screening participants in Huadu District,Guangzhou City.The case group included individuals diagnosed with T2DM during the screening,while the control group was matched from non-T2DM individuals according to the principles of age difference within 5 years and the same gender.Basic information about the study participants was collected using questionnaires.The expression levels of ncRNAs in serum were measured using the quantitative real-time PCR(qRT-PCR)method.A mediation model was applied to analyze the relationships among miRNA,lncRNA,and circRNA with T2DM,aiming to explore the impact of their expression levels on the development of T2DM.Results The case group and the control group each included 101 participants.The relative expression level of serum hsa_circ_0071106 in the case group was 1.175(0.701,1.748),while in the control group,it was 1.002(1.001,1.009).The serum hsa_circ_0071106 showed significantly higher expression in the case group(W=1 679,P=0.004).The relative expression level of serum hsa-miR-607 in the case group was 1.313(0.783,1.928),compared to 1.020(1.010,1.030)in the control group,with a statistically significant difference between the two groups(W=2 421,P<0.001).The relative expression level of serum lncRNA MEG3 in the case group was 1.381(0.605,2.629),while in the control group,it was 1.008(1.002,1.027),showing a statistically significant difference between the two groups(W=2 221,P<0.001).The results of the logistic regression model demonstrated that for each unit increase in hsa_circ_0071106,hsa-miR-607,and lncRNA MEG3 in the case group,the risk of developing T2DM increased by 1.839 times(95%CI=1.529~5.273,P=0.001),3.169 times(95%CI=2.078~8.362,P<0.001),and 1.088 times(95%CI=1.381~3.156,P<0.001),respectively.After adjusting for confounding factors such as low-density lipoprotein,the risk of developing T2DM increased by 1.958 times(95%CI=1.220~7.170,P=0.016)for each unit increase in hsa_circ_0071106.The mediation analysis revealed that using hsa-miR-607 as the mediating variable,the mediating effect of hsa_circ_0000284 on T2DM was statistically significant,with a 95%confidence interval of 0.000 7~0.127 0.Conclusion The elevated expression levels of hsa_circ_0071106,hsa_circ_0000284,hsa-miR-29a-5p,hsa-miR-3690,hsa-miR-607,lncRNA MEG3,and lncRNA TUG1 are associated with an increased risk of developing T2DM.Among these,hsa-miR-607 acts as a mediating variable and influences the expression of hsa_circ_0000284 in the pathogenesis of T2DM.关键词
