猪副流感病毒5型的分离鉴定及进化分析

Abstract
In order to investigate the genetic variation of porcine parain fluenza virus 5.PIV5,the NP gene,F gene and HN gene of PIV5 strains from Heilongjiang,Fujian and Hunan were sequenced and analyzed,respectively,to clarify the relationship between genome and genetic evolution.The three isolated strains were named DDB-P0,FJCT-P6 and HUNTX-P6,and five pairs of primers were designed with reference to the PIV5 gene sequence in Genbank.RT-PCR fragment amplification and sequencing were performed on the three PIV5 strains,and the gene sequences were analyzed for homology and genetic evolution.The results showed that the genome length of NP gene,F gene and HN gene of DDB-P0,FJCT-P6 and HUNTX-P6 strains was 1 530 nt,1 656 nt and 1 698 nt.The highest homology of the three strains was SH/2015/1202 strain from China,which was 99.9%.The distant relative was D277 strain of dog origin from Korea.Phylogenetic analysis showed that the three strains were closely related to SH/2015/1202 from Chinese pigs.This study provides a reference for the follow-up study of PIV5.关键词
