摘要

Abstract

[Objective]The pathogenic fungi infecting Lentinula edodes stick were isolated and identified to provide a theoretical basis for cultivation management and disease control.[Method]Diseased samples of rotting L.edodes were collected from cultivation greenhouses.Pathogens were isolated using the single-spore isolation method.Phylogenetic trees were constructed based on traditional morphological characteristics and rDNA ITS sequencing.Pathogenicity of the isolates was verified via Koch's postulates to confirm the fungal species responsible for the rot.[Result]Three fungal isolates(2023X110302-4,2023X110301-2 and 2023X110301-4)were obtained from the rotting samples.rDNA ITS sequencing revealed>99.83%homology with Trichoderma sp.TAMA 159 and>99.65%homology with T.citrinoviride HS13.However,significant morphological and cultural differences were observed compared to these reference strains.Pathogenicity tests confirmed that the Trichoderma isolates caused green mold disease in L.edodes.[Conclusion]The pathogen responsible for rot in L.edodes stick was identified as a Trichoderma species.Despite high genetic homology(>99.65%)with Trichoderma sp.TAMA 159 and T.citrinoviride HS13,the isolates exhibited distinct cultural and microscopic characteristics.Combined with phylogenetic analysis,the isolates likely represent a novel species within the Trichoderma genus.
