超声波辅助离子液体提取革命菜总黄酮的工艺优化及抗氧化活性研究
Optimization of Ultrasonic-assisted Ionic Liquid Extraction of Total Flavonoids from Gynura crepidioides and Its Antioxidant Activity

Abstract
To improve the resource utilization rate and increase the added value of Gynura crepidioides,total flavonoids were extracted from Gynura crepidioides and their antioxidant activity was evaluated.Ultrasonic-assisted ionic liquid technology was used to improve the extraction yield of total flavonoids.The effects of four key factors(ultrasonic temperature,ultrasonic time,ionic liquid molar concentration and liquid-material ratio)on the extraction yield were investigated.Based on the single factor experiment,Box-Behnken response surface model was further introduced to optimize the extraction process.The results showed that the best extraction effect can be achieved under the conditions of ultrasonic temperature of 74℃,ultrasonic time of 31min,ionic liquid molar concentration of 0.62mol/L and liquid-material ratio(mL/g)of 30∶1.Under these conditions,the actual extraction yield reached 59.30mg/g,and the relative error with the predicted maximum value was only 0.27%,which verified that the regression model had high prediction accuracy and reliability for the extraction process.Compared with hot extraction,the extraction yield of total flavonoids by this method increased by 86.01%.In addition,the total flavonoids of Gynura crepidioides showed certain potential in scavenging DPPH·and·OH,and their median inhibition concentration(IC50)values were 64.45 mg/L and 150.37 mg/L,respectively.This study showed that the total flavonoids of Gynura crepidioides had certain antioxidant activity,which provided scientific basis for the deep development and comprehensive utilization of Gynura crepidioides resources.关键词
