摘要

Abstract

This research was conducted to establish a specific and sensitive PCR method to detect Apicystis bombi.A pair of specific primers based on the conserved sequences of the ITS genes of A.bombi were designed.After optimizing the reaction conditions and system,a PCR detection method for detecting A.bombi was developed,and the specificity,sensitivity and stability of the method were tested.The results showed that the detection limit of this method for detection of A.bombi was 2.19×10-5 ng/μL,and there was no cross-reaction with other 5 parasites,including Critidia bombi,Nosema bombi,Locustacarus buchneri,Nosema apis and Nosema cerana.The method had good sensitivity and specificity,and showed good stability at the level of the detection limit.The detection results of 57 clinical samples detected by the method were consistent with the sequencing results.The detection method developed in this study could be applied to quarantine,laboratory testing and epidemic monitoring of A.bombi.
