Abstract

In order to study the biological characteristics of β-interferon(IFN-β)gene in Aonyx cinerea,based on the β-interferon(IFN-β)gene sequence of Enhydra lutris and Mustla vison published in GenBank,we designed and synthesized a pair of specific primers for the amplification of otter IFN-β gene.At the same time,bioinformatics analysis of IFN-β was performed.The results showed that the gene length of the Asian claw otter IFN-β was 561 bp,coding a total of 186 amino acids,the protein relative molecular mass was 22.07 kDa,the electric point was 6.43,the molecular formula could be expressed as C997H1573N259O289S8,and the total number of atoms was 3 126.The protein is a stable hydrophilic protein,the first 21 amino acids are signal peptides,and has a transmembrane structure.Amino acids 1-4 are located outside the membrane,amino acids 5-27 in the transmembrane region,and amino acids 28-186 in the membrane.The obtained IFN-β gene of Asiatic otter was compared by homology and genetic evolution analysis,the IFN-β gene of Aonyx cinerea was the closest to that of Enhydra lutris published in GenBank(XM_022517563.1),and the nucleotide similarity are 99.3%.The evolutionary tree showed that the IFN-β gene of Aonyx cinerea belonged to a small branch with that of Enhydra lutris and Lontra canadensis,and the kinship is the closest.The results laid a foundation for the subsequent study on the protein expression and biological activity of the otter IFN-β gene.
