Abstract

Rapid and accurate identification of pigeon sex is one of the most important tasks to increase pigeon breeding rate and economic benefit.Five traditional methods were used to identify the sex of 100 4-month-old pigeons.At the same time,molecular biology techniques were used,including chromobox helicase DNA binding gene(CHD gene)specific primers for PCR amplification,sequencing and nucleotide homology analysis.And the comparison between the traditional identification methods and PCR method was conducted.The results showed that in traditional avian sexing methods,the accuracy rates were determined as follows:vent sexing demonstrated 57%accuracy,vocalization analysis achieved 52%,skeletal morphology assessment reached 48%,plumage characteristics evaluation attained 45%,and morphometric analysis(body size evaluation)showed 47%accuracy.Notably,the integrated approach combining vent sexing,vocalization analysis,and skeletal morphology assessment yielded the highest diagnostic efficacy with an 86%accuracy rate,the PCR method could improve the accuracy of sex identification of pigeon rapidly and accurately compared with the traditional methods.This experiment provides an accurate method for sex identification of pigeons and plays an important role in improving the economic benefits for pigeon farm.
