马铃薯无糖组织培养的条件优化及其在马铃薯原原种生产中的应用
Optimization of sugar-free tissue culture conditions for potato and its ap-plication in pre-basic seed production

Abstract
To further optimize the microenvironment parameters of sugar-free tissue culture,this study used Qingshu No.9 as the test material and designed different gradient experiments with different ratios of vermiculite to nutrient solution(Vvermiculite∶Vnutrientsolution=1.0∶0.7,Vvermiculite∶Vnutrient solution=1.0∶0.8,Vvermiculite∶Vnutrient solution=1.0∶0.9),CO2 concentrations(500 μmol/mol,800 μmol/mol,1 100 μmol/mol,1 400 μmol/mol),and light intensities to investigate the effects of each treatment on the growth and development of potato tissue-cultured seedlings.The light intensity was set at 50 μmol/(m2·s),75 μmol/(m2·s),100 μmol/(m2·s).The results showed that the optimal conditions for sugar-free tissue culture of po-tatoes were as follows:Vvermiculite∶Vnutrient solution=1.0∶0.8,CO2 concentration of 800 μmol/mol,and light intensity of 75 μmol/(m2·s).Compared with traditional tissue culture,the yield,quantity,and survival rate of potato pre-basic seeds obtained through sugar-free tissue culture were significantly increased by 16.09%,22.37%,and 3.27%,respectively(P<0.05).In conclusion,this study not only clarified the key technical parameters of sugar-free tissue culture but also confirmed the advantages of this technology in improving the production efficiency of potato seed tubers,providing theoretical basis and techni-cal support for the standardized production and industrial propagation of potato tissue-cultured seedlings.关键词
