锰离子胁迫下解淀粉芽孢杆菌A-3代谢响应
Metabolic response of Bacillus amyloliquefaciens A-3 under manganese ion stress

Abstract
In order to study the metabolic response of Bacillus amyloliquefaciens A-3 to Mn2+stress,a sterile fermentation broth of A-3 was used as the research object.All metabolites of A-3 were identified by non-targeted metabolomics technology by adding Mn2+.According to the importance of variable weights(VIP>1),the differentially expressed metabolites were screened by T-test(P<0.05),and the A-3 metabolic pathway was enriched by KEGG database.The results showed that Mn2+stress had a significant effect on the changes of metabolites in A-3 fermentation broth.A total of 332 differential metabolites were identified,175 were detected in positive ion mode and 157 were detected in the negative ion mode.Among them,114 metabolites were up-regulated,such as 3-methylindolepyruvate,spermine,adenosine and so on.Down regulation of 218,such as 5-hydroxylysine,citric acid,homogeneous acid and other small analytical organic acids.Through KEGG enrichment and topological analysis of metabolic pathways,22 metabolic pathways with P<0.05 were found,among which the metabolic pathways of alanine,aspartate and glutamate were most significantly affected.The addition of Mn2+changes the metabolic level of A-3 cells,among which the expression of spermine is up-regulated.Spermine was involved in glutathione metabolism,pantothenate and CoA biosynthesis and other important metabolic and biosynthetic pathways.A-3 mainly responded to Mn2+stress by regulating the expression of amino acids,nucleosides and small molecular organic acids.关键词
