摘要

Abstract

Objective Recurrent chronic inflammation in ulcerative colitis(UC)may cause cancer.This study observed the protective effect of mild moxibustion and electroacupuncture(EA)on the intestinal mucosa of UC rats and the immunoregulatory mechanism.Method Sprague-Dawley(SD)rats were randomized into a normal group,a model group,a mild moxibustion group,an EA group,and a Western medicine group.The UC model was established by intraperitoneal injection of Azoxymethane(AOM)(10 mg/kg)plus freely drinking 5%Dextran sulfate sodium salt(DSS)solution for the subsequent 5 consecutive days.The general condition,colon length,and colonic histopathology and score were observed.The enzyme-linked immunosorbent assay(ELISA)was adopted to detect the serum contents of interleukin-6(IL-6)and transforming growth factor beta 1(TGF-β1);the immunofluorescence(IF)technique was used to locate 5-methylcytosine(5-mC),5-hydroxymethylcytosine(5-hmC),ten-eleven translocation(TET)family proteins,and histone deacetylase 2(HDAC2)and detect their expression in the colonic tissue.Result Compared to the normal group,the model group had a significantly shorter colon length and an increased colonic histopathological score(P<0.05),along with an elevated IL-6 content and a decreased TGF-β1 content in the serum(P<0.05),increased 5-mC protein expression(P<0.05),and reduced protein expression of 5-hmC,TET1,TET2,TET3,and HDAC2(P<0.05).Compared to the model group,the mild moxibustion,EA,and Western medicine groups all demonstrated an increased colon length and a declined pathological score(P<0.05),together with an elevated TGF-β1 content and a decreased IL-6 content in the serum(P<0.05),increased protein expression of 5-hmC,TET1,TET2,TET3,and HDAC2 in the colonic tissue(P<0.05);the mild moxibustion group also showed decreased colonic 5-mC protein expression(P<0.05).In the mild moxibustion group,TET1,TET2,and TET3 were found co-expressed with HDAC2 in the colon;TET2 and TET3 co-expressed with HDAC2 in the EA group;TET1 and TET3 were found co-expressed with HDAC2 in the Western medicine group.Conclusion Both mild moxibustion and EA can produce protective effects on colonic mucosa in UC,and the mechanism should be related to their effects on up-regulating the protein expression of DNA hydroxymethylases TETs and HDAC2 and maintaining the 5-mC/5-hmC balance.Further,mild moxibustion can recruit HDAC2 via TET1,TET2,and TET3,while EA only via TET2 and TET3,to play the immunoregulatory role.
