Abstract

Microorganisms are an important component of atmospheric bioaerosols,which can be re-moved from the air by wet deposition.Therefore,a large number of microorganisms,such as bacteria,are loaded in rainwater.In this study,a rainwater collector was used to continuously collect samples in the coastal region of Qingdao from December 2020 to December 2022.Meanwhile,viable/non-viable bacteria concentrations were measured by using fluorescence microscopy after staining with BacLightTM bacterial viability kit.The variation of concentration,distribution characteristics and influencing factors were analyzed.The results showed that during the sampling period,the concentration of viable and non-viable bacteria in rainwater was(4.70±4.63)×102 and(3.32±2.21)× 103 cells·mL-1,respective-ly,which varied greatly depending on different precipitation.The concentration of non-viable bacteria was the highest in spring,lower in summer and autumn,and the lowest in winter,while the concentra-tion of viable bacteria was the highest in winter,lower in autumn and summer,and the lowest in spring.The concentration of bacteria in rainwater showed monthly variations,with the highest average concentration of viable bacteria appearing in January and August;while the highest monthly average concentration of non-viable bacteria appearing in June.The monthly distribution was related to the source and distribution of bacteria in the air,environmental condition,weather condition,and so on.The viability of bacteria in rainwater ranged from 0%~47.42%,with an average of 12.4%,which showed seasonal differences.Correlation analysis showed that the concentration of viable bacteria and bacteria viability in rainwater were negatively correlated with wind speed(p<0.01),but had no corre-lation with rainfall and air quality indexes(p>0.05).The concentration of total and dead bacteria were negatively correlated with K+(p<0.05).The source of air mass also affected the microorganisms con-centration in rainwater.Backward trajectories analysis showed that the concentration of viable bacteria and bacteria viability in rainwater,affected by marine sources,are higher than those from land sources,indicating that the ocean had an impact on the concentration of bacteria in rainwater in coastal areas.
