Abstract

Objective:To screen cytotoxic T lymphocyte(CTL)-efficiently recognized antigenic epitopes in Coxsackievirus B3(CVB3)and provide a theoretical basis for developing broad-spectrum vaccines against CVB3 and other enteroviruses.Methods:CTL candidate epitopes of CVB3 were predicted using the Immune Epitope Database(IEDB)and NetMHCpan EL 4.1 algorithm.For HLA-A*02:01-restricted epitopes,AlphaFold 2.3.2 was employed to simulate their complex structures with human leucocyte antigen(HLA),analyzing epitope conservation and cross-immunity potential.The affinity of candidate epitopes to HLA-A*02:01 was validated through T2 cell binding assays.Results:A total of 71 potential CTL epitopes were identified,with 6 high-affinity HLA-A*02:01-restricted epitopes(prediction score>0.9)selected for further analysis.In vitro experiments confirmed that 5 epitopes(CTL1-5)significantly induced HLA-A*02:01 expression(P<0.05),while CTL6 showed no binding activity.Conservation analysis revealed that CTL1-6 epitopes were highly conserved across Coxsackievirus,Echovirus,and Enterovirus strains,suggesting their cross-protective potential.Conclusion:This study identifies HLA-A*02:01-restricted CTL epitopes in CVB3,providing critical targets for the development of broad-spectrum vaccines against CVB3 and related enteroviruses.
