超低温保存对3个品种海岛棉种子活力及幼苗生理指标的影响
Effects of cryopreservation on seed viability and seedling physiological indexes of three Gossypium barbadense L.varieties

Abstract
In this experiment,three varieties of Gossypium barbadense L.seeds were used,namely'Haipinbi II''Xinhai 14'and'504-V',and the method of cryopreservation was adopted.After 24 h of cryopreservation,the germination rate and germination potential of the seeds were observed after different freezing temperatures,as well as the changes in peroxidase(POD)activity,superoxide dismutase(SOD)activity,catalase(CAT)activity,malondialdehyde(MDA)content,and photosynthetic pigment content in the seedling leaves.It was found that the germination rate and germination potential of'Haipinbi II'were lower than'Xinhai 14'and'504-V'before and after ultra-low temperature treatment.After cryopreservation,the optimal freezing temperatures for germination of'Haipinbi II''Xinhai 14'and'504-V'were 25℃,55℃,and 65℃,while the lowest freezing temperatures were 35℃,25℃,and 35℃,respectively.Overall,POD activity,SOD activity,CAT activity,MDA content,and photosynthetic pigment content showed a trend of initially decreasing,then increasing,and then decreasing with the increase in freezing temperature.The results showed that it was feasible to preserve the seeds of these three varieties of Gossypium barbadense L.at cryopreservation without any treatment,and the survival rates of'Haipinbi II''504-V'and'Xinhai 14'were 71.11%,96.67%and 97.78%,respectively,which provided a theoretical basis for the conservation of Gossypium barbadense L.germplasm resources.关键词
