柴胡皂苷A体外抑制猪轮状病毒增殖的研究
Saikosaponin A inhibits porcine rotavirus replication in vitro

Abstract
In vitro screening of the bioactive component of traditional Chinese medicine with antiviral effects on porcine rotavirus(PoRV)replication provided a theoretical guide for the development of antiviral drugs and the prevention of PoRV infection.The potential anti-PoRV activity of Saikosaponin A(SSA)was identified from six active ingredients of Chinese medicine.The cytotoxicity of SSA on IPEC-J2 cells was determined using a CCK8 assay.IPEC-J2 cells were treated with different concentrations of SSA and were then infected with PoRV NJ2012 strain.The replication levels of PoRV were analyzed by RT-qPCR,TCID50 and Western-blot assays.Furthermore,the effect of SSA on PoRV replication cycle was investigated.Finally,the effect of SSA on the propagation of diverse genotypes of PoRV(G9P[23]、G1P[7]and G4P[23]),simian RV,and bovine RV was examined.The results of the CCK8 assay showed that SSA had no cytotoxic effect on the IPEC-J2 cells under the concentration of 120 μg/mL.SSA inhibited PoRV NJ2012 strain propagation in a dose manner with different concentrations.Further studies demonstrated that SSA decreased PoRV propagation at the replication stage,while having no significant effect on the stages of binding,entry,and release.Additionally,SSA suppressed the replication of different genotypes of PoRV,simian RV,and bovine RV to varying degrees.In summary,this study demonstrated that SSA could significantly inhibit PoRV propagation at the replication stage in vitro.SSA also exhibited broad-spectrum anti-RV activity against diverse genotypes or species of RV,suggesting its potential for the development of antiviral drugs to control PoRV spread in pigs.关键词
