Abstract

This study aimed to express the H protein of peste des petits ruminants virus(PPRV)as well as to predict and identify its linear B cell epitopes.The H gene of PPRV was amplified by PCR,and then it was used to construct the recombinant plasmid pET-30a(+)-H.The H protein expressed by the prokaryotic expression system was purified and refolded.Then the refolded H protein was used to immunize mice to prepare polyclonal antiserum.The linear B cell epitopes of H protein were predicted and analyzed by bioinformatics software.The nucleotide sequences of 16 potential epitopes among those predicted were respectively recombined into the pGEX-4T-1 vector.The fusion expression of the predicted epitopes and GST tag protein was accomplished by prokaryotic expression system.The purified fusion proteins of the predicted epitopes were identified by Western-blot and indirect ELISA with the utilization of the prepared polyclonal antiserum.The results showed that the H protein had good immunogenicity.Five linear B cell epitopes were screened and identified by the recombinant H protein polyclonal antibody.This study successfully expressed the H protein of PPRV and predicted and identified five linear B cell epitopes.It provides a reference for the development of diagnostic reagents and epitope vaccines for peste des petits ruminants.
