Abstract

Gelatin microspheres were prepared using Ophiopogon japonicus fibrous root extracts as the active ingredient and gelatin as the carrier through an emulsification-crosslinking method.The effects of the amount of hardener,emulsifier concentration,the gelatin mass fraction and water-oil ratio on the encapsulation rate and drug loading capacity of the microspheres were investigated.The optimal preparation prescription was determined by orthogonal test.The morphology of gelatin microspheres loaded with extract of Ophiopohon japonicus fibrous roots was observed by scanning electron microscope(SEM).Paddle method was applied to in vitro release test of gelatin microspheres.In addition,the curve of drug release in vitro was fitted with different release model to analyze the in vitro release characteristics of gelatin microspheres loaded with extract of Ophiopohon japonicus fibrous roots in nasal cavity,synthetically.The stability of gelatin microspheres loaded with extract of Ophiopohon japonicus fibrous roots was investigated under the conditions of constant temperature and humidity at 40 ℃ and 75%RH for 60 days.The results showed that the optimal prescription was as follows:the amount of hardener of 7%,emulsifier concentration of 15%,the gelatin mass fraction of 4%and water-oil ratio of 1∶8,and the encapsulation rate and drug loading capacity were 69.42%and 15.04%respectively.Under the SEM,the microspheres appeared round,the surface was smooth and the average particle size was 64.5 μm.The release curve of erythromycin gelatin microspheres in vitro could be appropriately simulated by Higuchi equation.In the stability study,the retention rate of gelatin microspheres was 96.1%.These findings confirmed that the preparation process for gelatin microspheres loaded with extract of Ophiopohon japonicus fibrous roots was stable and practical,exhibiting good sustained-release characteristics.It provides a theoretical basis for future product development.
