Abstract

In order to obtain the entomogenic fungi with high pathogenicity to the larva of Clostera anachoreta in the high cold area,the entomogenic fungi were isolated from the cadavers of adult spruce-flowered longhorn beetle in the Weihe Town Forest Farm of Harbin City in Heilongjiang Province.The entomopathogenic fungi were identified through morphological observation and molecular biological analysis,and then their virulence to the larva of the tox-icity test and extracellular enzyme activity were determined.The results showed that the isolated and purified strain DU01 was Beauveria bassiana.Under the infection of strain DU01 with different concentration of spore suspension(1×108,1×107,1×106,1×105,1×104 spores/mL),the mortality of the larva was increased with the increase of the concentration of spore suspension.When the concentration was 1.0×108 spores/mL,the time to death of larva was 3.84 d,and the adjusted mortality of larva was 100%after 7 days of inoculation.Protease activity and chitinase activity reached the maximum value on the 4th day,5.65 U/mL and 4.77 U/mL,respectively.The results indicated that Beauveria bassiana had strong pathogenicity to the larva of Clostera anachoreta,which could provide a new idea for the green control of Clostera anachoreta.
