CpG-ODN Nanoparticle Enhances Mucosal Immunity and Its Protective Efficacy Against Avian Pathogenic Escherichia coli

Abstract
To develop CpG oligodeoxynucleotide(CpG-ODN)nanoparticles that enhance mucosal immunity and provide support for antibiotic reduction and alternatives,this study prepared CpG-ODN nanoparticles using multi-walled carbon nanotube(MWCNT).The nanoparticles were administered via the intrapulmonary route to newly hatched Hy-Line W-36 white-shell laying hens.Enzyme-linked immunosorbent assay(ELISA)was used to measure levels of interleukin-2(IL-2),interferon-γ(IFN-γ)in serum,and intestinal secreted immunoglobulin A(sIgA).Quantitative real-time reverse transcription polymerase chain reaction(qRT-PCR)was employed to detect mRNA expression levels of interleukin-1β(IL-1β),interleukin-4(IL-4),interleukin-6(IL-6),interleukin-8(IL-8),interleukin-10(IL-10),interleukin-18(IL-18)and Toll like receptor 21(TLR21).The protective efficacy against Escherichia coli O2 challenge was evaluated by survival rate,clinical scoring,and bacterial colonization scores.The results showed that the average particle size of the CpG-ODN nanoparticles was(72.30±9.54)nm with uniform morphology.Compared with the Control group,IL-2 and IFN-γ levels in serum were significantly elevated at 12 and 24 hours post-immunization(P<0.01).mRNA levels of IL-1β,IL-6,IL-18,IL-4,and TLR21 were significantly increased at 12 and 24 hours(P<0.05 or P<0.01);IL-10 mRNA expression was significantly increased at 6 hours(P<0.01)and moderately increased at 12 hours(P<0.05).Intestinal sIgA levels were significantly elevated at 12 and 24 hours(P<0.05)and highly significantly at 48 hours(P<0.01).Compared to the Challenged control group,both the CpG-ODN and Antibiotic-treated groups showed significantly higher survival rates(P<0.05),and significantly lower clinical and bacterial colonization scores(P<0.05).These findings indicate that intrapulmonary administration of CpG-ODN nanoparticles can activate mucosal immune responses,effectively protect chickens from challenge with avian pathogenic E.coli O2,alleviate clinical symptoms,and reduce mortality.The results demonstrate promising potential for CpG-ODN nanoparticles as antibiotic alternatives or supplements.关键词
