摘要

Abstract

To assess the prevalence of Theileria sinensis(T.sinensis)infection in yaks in parts of Qinghai Province,this study collected 411 blood samples from yaks across 10 counties in 6 prefectures.Genomic DNA was extracted from the blood samples,and nested PCR was used to amplify the V4 hypervariable region of the Piroplasm 18S rRNA gene for sequencing and analysis.The software DnaSP v6 was used to analyze the number of haplotypes,mutation sites,haplotype diversity,and nucleotide diversity of the positive samples.PopArt and MEGA 6.0 software were used to construct a haplotype network and phylogenetic tree,respectively.The results showed a T.sinensis infection rate of 19.46%(80/411)among yak blood samples.A total of 16 haplotypes of the T.sinensis 18S rRNA gene were identified,with Theileria sinensis haplotype 1(TsH1)being dominant(65/80),found across Haiyan,Henan,Dari,Guinan counties,and Yushu City.The remaining 15 haplotypes each accounted for 1.25%(1/80)and were exclusively found in Guinan County.The haplotype diversity index was 0.342,and nucleotide diversity was 0.002 53,with a total of 25 mutation sites.Phylogenetic analysis revealed that the 16 haplotypes clustered with previously published T.sinensis strains from Gansu,Wuhan,Hainan,Xinjiang,and Henan in GenBank.This study provides the essential data for the prevention and control of tick-borne T.sinensis infection in Qinghai Province.
