柔嫩艾美耳球虫湖南株的分离和鉴定
Isolation and Identification of Eimeria tenella Hunan Strain

Abstract
To identify the Eimeria species responsible for mortality in 6-week-old broilers at a yellow-feather broiler farm in Changsha,Hunan Province,this study successfully isolated a coccidian strain from feces and cecal contents of dead chickens using saturated salt flotation and single oocyst isolation techniques.Preliminary identification was performed by observing the morphology and size of the oocysts.The species was further confirmed by polymerase chain reaction(PCR).Biological characteristics of the strain were subsequently assessed,including prepatent period,site and size of schizonts,pathogenicity,and minimum sporulation time.The results showed that the strain parasitized the cecum,with average oocyst size of 22.2 µm×19.0 µm and an oocyst shape index of 1.17.Sequence alignment of the PCR-amplified product indicated 96.44%-100%nucleotide homology with seven known strains of Eimeria tenella listed in GenBank.The strain was designated as E.tenella-TCS2404-01.Biological characterization revealed a prepatent period of 124 hours,a minimum sporulation time of 16 hours,and inoculation of 1.2×10⁴ oocysts per bird resulted in a relative weight gain reduction to 73.8%,an average intestinal lesion score of 3.7,and a mortality rate of 30%.These findings confirm the isolation of a highly pathogenic E.tenella strain.This discovery is of significance for understanding the epidemiology and control of coccidiosis in Hunan Province and provides foundational data for future vaccine development and drug research targeting this strain.关键词
