摘要

Abstract

In order to establish a high-throughput and fully automated magnetic particle chemiluminescent immunoassay(MP-CLIA)for detecting antibodies against foot-and-mouth disease virus serotype O(FMDV-O),a magnetic particle chemiluminescence method for FMDV-O was established through optimizing all reaction conditions using FMDV-O-VP1 structural protein and enzyme-labelled monoclonal antibody.The results showed that the antibodies against FMDV-O were detected positive and negative when the critical values(S/N)were≤0.8 and>0.8,respectively.The established method was with good specificity without cross-reaction with immune sera of five common pathogens;and its limit of detection(LOD)for sensitivity control serum was 1:1 024,and intra-and inter-assay coefficients of variation(CVs)were within 10%,which was 95.9%compliant with the standard method.In conclusion,the established method presented high sensitivity,specificity and reproducibility,and was an efficient method for detecting immune antibodies against FMDV-O.
