化脓隐秘杆菌灭活疫苗佐剂的筛选与免疫保护性评价OA
Screening of Adjuvants for Inactivated Vaccine of Arcanobacterium pyogenes and Evaluation on Their Immunoprotective Efficacy
为筛选适用于化脓隐秘杆菌灭活疫苗的合适佐剂,分别将化脓隐秘杆菌抗原与4种佐剂混合,制备了4种灭活疫苗(ISA 15佐剂疫苗、ISA 61佐剂疫苗、ISA 201佐剂疫苗和氢氧化铝佐剂疫苗).随后,对4种化脓隐秘杆菌灭活疫苗进行物理性状观察和安全性检验,并将疫苗免疫小鼠,以检测血清抗体水平及疫苗免疫保护效果.结果显示:4种佐剂疫苗物理性状均合格;注射小鼠后,4种佐剂疫苗均有副反应(小鼠背部红肿),其中ISA 61疫苗的副反应最轻微;ISA 61组小鼠血清抗体水平极显著高于其他组(P<0.01),其次是ISA 201组;三免后7 d,使用化脓隐秘杆菌对小鼠攻毒,发现对小鼠保护率最高的为ISA 61佐剂疫苗,保护率达90%,其次为ISA 201、ISA 15、氢氧化铝佐剂疫苗以及无佐剂灭活疫苗,保护率依次为60%、50%、40%、30%.结果表明,ISA 61佐剂疫苗副反应较小,诱导产生的抗体水平最高,同时提供了最高保护效力,即ISA 61为制备化脓隐秘杆菌灭活疫苗的首选佐剂.本研究为进一步开发化脓隐秘杆菌灭活疫苗奠定了基础.
In order to select an adjuvant appropriate for inactivated vaccine of Arcanobacterium pyogenes,4 kinds of inactivated vaccines(including ISA 15 adjuvant vaccine,ISA 61 adjuvant vaccine,ISA 201 adjuvant vaccine and aluminum hydroxide adjuvant vaccine)were prepared using 4 adjuvants,respectively,which were observed for physical properties and tested for safety,and then immunized into mice to detect serum antibodies and the immunoprotective efficacy.The results showed that all the 4 adjuvant vaccines were qualified in physical properties,but caused adverse reaction(dorsal inflammation)after injection,which was the slightest in mice immunized with ISA 61 adjuvant vaccine;serum antibody level of mice in ISA 61 group was extremely significantly higher than that of other groups(P<0.01),followed by mice in ISA 201 group;7 days after the third immunization,mice were challenged with Arcanobacterium pyogenes,the protection rate provided by ISA 61 adjuvant vaccine was the highest,reaching 90%,followed by ISA 201,ISA 15,aluminum hydroxide adjuvant vaccine and adjuvant-free inactivated vaccine,remaining 60%,50%,40%and 30%,successively.In conclusion,ISA 61 adjuvant vaccine could cause less side effects,induce antibody level at the highest level,and provide the highest protective efficacy,which was preferred for preparing inactivated vaccine of Arcanobacterium pyogenes.A foundation was laid by the study for further development of inactivated vaccine of Arcanobacterium pyogenes.
