艳山姜纯露对脂多糖致兔肝脏、肺脏和下丘脑损伤的保护作用及机制
Protective Effects and Mechanisms of Alpinia zerumbet Hydrosol against LPS-Induced Liver,Lung and Hypothalamic Indury in Rabbits

Abstract
This study aimed to investigate the protective effect and mechanism of Alpinia zerumbet hydrosol on lipopolysaccharide(LPS)-induced injury in the liver,lung,and hypothalamus of rabbits.A total of 36 male New Zealand white rabbits were randomly divided into six groups:Control group,Model group,Ibuprofen group,and Low-,Medium-,and High-dose A.zerumbet hydrosol groups.The Low-,Medium-,and High-dose groups received A.zerumbet hydrosol via gavage at doses of 1.8,3.6,and 7.2 g/(kg·bw·d),respectively,for 7 consecutive days.The Control and Model groups were administered an equal volume of purified water,while the Ibuprofen group was given a single dose of ibuprofen at 10 mg/(kg·bw·d).Thirty minutes after the last administration,rabbits(except those in the Control group)were injected with LPS via the marginal ear vein at a dose of 4 µg/(kg·bw)to induce fever.Rectal temperature was recorded at 0,1,3,and 6 h after LPS injection.At 6 h post-injection,ELISA was used to detect serum levels of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),interleukin-6(IL-6),and prostaglandin E2(PGE2).Pathological changes in the liver,lung,and hypothalamus were observed by hematoxylin-eosin(H.E.)staining.The expression of cyclooxygenase-2(COX-2)in the liver,lung,and hypothalamus,and Toll-like receptor 4(TLR4)and nuclear factor-κB p65(NF-κB p65)in the hypothalamus,were detected by immunohistochemistry.The results showed that the LPS-induced fever and organ injury model was successfully established.Compared with the Model group,all indices in the Ibuprofen group improved significantly(P<0.05).Rectal temperatures in the Low-dose group(at 6 h),Medium-dose group(at 3 and 6 h),and High-dose group(at 1,3,and 6 h)were significantly reduced(P<0.05).Serum IL-6 and PGE2 levels were significantly decreased in all A.zerumbet hydrosol groups(P<0.05),and TNF-α and IL-1β levels were significantly reduced in the Medium-and High-dose groups(P<0.05).Histopathological injury in the liver,lung,and hypothalamus was notably alleviated,and COX-2 expression in these tissues,as well as TLR4 and NF-κB p65 expression in the hypothalamus,were significantly downregulated(P<0.05).These findings suggest that A.zerumbet hydrosol confers protective effects against LPS-induced liver,lung,and hypothalamic injury in rabbits,potentially through reducing endogenous pyrogens and inhibiting the expression of TLR4,NF-κB p65,and COX-2.This study provides a theoretical foundation for the veterinary clinical application of A.zerumbet hydrosol in alleviating fever and organ injury.关键词
