Abstract

In order to explore the optimal immunization conditions for the attenuated vaccine Sm24_Rif12_Ssq for Salmonella enteritidis,the age at first immunization,immunization dose and acidifier application interval were tested and analyzed.Hy-Line variety brown cocks(0-day-old)were selected and orally immunized with 1 avian-dose vaccine at 0 and 7 days of age.Cloacal swabs were collected 1 to 7 days after the immunization to detect Salmonella using qPCR,and to analyze the optimal age for the operation;cocks at 0-day-old were orally administered with 1.0 and 0.5 avian-dose vaccines,then their livers were collected 1 to 14 days after the immunizatio n for bacterial count,and cloacal swabs for Salmonella detection by qPCR to analyze the optimal dose;cocks at 0-day-old were orally administered with 1 avian-dose vaccine,and fed with feed containing acidifier at 0,3 and 7 days after the immunization,cloacal swabs were collected 1 to 15 days after the immunization to detect Salmonella by qPCR,and to analyze the optimal acidifier application interval.The results showed that the cloacal Salmonella positive rate of 0-day-old immunized cocks was from 30%to 85%,but that of 7-day-old immunized cocks was only from 0%to 20%;there was no obvious difference(P>0.05)in the liver bacterial load and cloacal Salmonella positive rate of cocks immunized with 1.0 and 0.5 avian-dose vaccines;the positive rate was not affected(P>0.05)by acidifier application at different intervals after the immunization.In conclusion,the vaccine for Salmonella enteritidis was suitable for the first immunization with 0.1 avian-dose vaccine at 0-day-old,and its intestinal colonization was not affected by acidifier application after the immunization.Data reference was provided for clinical administration of the attenuated vaccine Sm24_Rif12_Ssq for Salmonella enteritidis.
