摘要

Abstract

Porcine epidemic diarrhea virus(PEDV)infection causes severe diarrhea in piglets,leading to intestinal mucosal damage and immunodeficiency.In this study,the jejunal tissues of PEDV-infected piglets were collected for transcriptome sequencing analysis using RNA-Seq technology to explore the changes in genomic transcription level and screen differentially expressed genes in the host immune response.RNA-seq results showed that 339 significantly differentially expressed genes(DEGs)were identified in PEDV-infected piglets compared with normal uninfected piglets,including 182 up-regulated genes and 157 down-regulated genes.GO molecular function results showed that the immune-related differentially expressed genes were mainly related to the immunoglobulin and receptor binding,the T cell proliferation,etc.KEGG pathway enrichment results showed that the differentially expressed genes were mainly involved in TNF signaling pathway,cytokine-cytokine receptor interaction,toll-like receptor signaling pathway,Jak-STAT signaling pathway and RIG-I-like receptor signaling pathway.Finally,the differentially expressed genes were verified by qRT-PCR,and the results were basically consistent with the transcriptome results.This study provided a scientific basis for further study on the mechanism of changes in host immune response caused by PEDV infection.
