摘要

Abstract

In this study,Madin Darby canine kidney(MDCK)cells was used as the culture medium to amplify the Porcine influenza virus.Formaldehyde and β-Propiolactone(BPL)had been selected as chemical inactivators for virus inactivation.The optimal conditions for formaldehyde inactivation were 0.1%final concentration and 37℃ inactivation for 24 h.The optimal inactivation conditions for BPL were 0.02% final concentration and 4℃ inactivation for 18 h.The antigen hemagglutination titer was about one titer lower than the original titer after inactivation.On the basis of using different inactivating agents,the inactivated vaccine was emulsified with oil phase adjuvant ISA 201 VG and water phase adjuvant HA 208.BALB/c mice were immunized with inactivated vaccines to evaluate immune and antiviral protective effects,with a challenge dose of 1 ×106 TCID50 per mice.Formaldehyde inactivated vaccines could stimulate mice to produce high levels of HI antibodies compared to BPL inactivated vaccines.Higher antibody levels were also reflected in the use of adjuvants,that the oil phase adjuvant ISA 201 VG is more effective than the water phase adjuvant HA 208.The survival rate of the vaccine immunized group reached 100% after SIV challenged.By comparing the experimental data of the control group,the virus content in the lungs of immunized group was significantly reduced.The above results indicate that the inactivated vaccine with formaldehyde+ISA 201 VG adjuvant immunization of BALB/c mice could produce high level of antibodies and resist the attack of Eurasian avian-like SIV,providing a scientific basis for the research and production of inactivated SIV vaccines in the future.
