Abstract

Coenonia anatina is one of the important pathogenic bacteria in respiratory tract infections in watefowl,which causes clinical signs similar to those of exudative septicemia caused by Riemerella anatipestifer infection.However,few research reports are available in China regarding its culture,antibiotic resistance and pathogenicity.In this study,a C.anatina strain was isolated in Guangdong province from a goose with swelling of the suborbital sinus and identified through pathological dissection,16S rRNA determination and genetic evolution analysis.The growth and drug resistance of this strain were studied through proliferation curves,biochemical reactions and antibiotic susceptibility testing.The pathogenicity of this strain was observed by goose embryo infection.The bacterial culture showed a slight yellow,dew-drop like and round colonies with neat edges and smooth surfaces on the TSA medium post 48 h.The gene sequence analysis revealed that the 16S rRNA of this strain was 98.7% homologous with the representative strains of the genus C.anatina,suggesting they belonged to the same branch.This strain grew slower in TSB broth medium than other common bacteria.Biochemical test results showed no fermentation of sugar,alcohol and urea,negative hydrogen sulfide reaction,and positive oxidase and catalase reactions\.In addition,this strain was susceptible to a number of antibiotics but resistant to ampicillin,kanamycin,and rifampicin.Inoculation of this strain into goose embryos caused death from 1 dpi to 5 dpi,and the whole body of the dead embryos showed severe congested.The C.anatina bacteria were re-isolated from the yolk fluid of the dead goose embryos.In summary,this study was the first report of C.anatina isolation from a goose in China and the results of this study provided a reference for development of diagnosis and treatment measures and for monitoring and control of this disease in the future.
