摘要

Abstract

In order to find out the causative pathogens of the respiratory disease in a cattle farm in Jiangsu,30 nasal swab samples were collected,and the main pathogens were detected by RT-PCR/PCR methods.For virus-positive samples,sequencing and genetic evolution analysis were conducted.For Mycoplasma bovis(M.bovis)positive samples,M.bovis was isolated,biochemical experiments,growth curve determination,antimicrobial susceptibility tests and genetic evolution analysis were carried out on the isolates.The results showed that 18 samples were positive for M.bovis,1 was positive for Mannheimia haemolytica,2 were positive for Bovine viral diarrhea virus(BVDV)and 6 were Bovine coronavirus(BCoV)positive.All other pathogens were negative.The growth titers reached the peak at 12 hours post inoculation,and there was no significant difference in the growth curves of the M.bovis isolates.The isolates did not ferment glucose and mannitol,did not hydrolyze gelatin and esculin,did not decompose urea.Antimicrobial susceptibility tests showed the 3 M.bovis isolates were susceptible to tymycin,lincomycin,and tywanectin.The sequence homology of the oppF gene of the isolates was 98.6%-100%with the reference strains,and they were closely related to the domestic strains.BVDV 5'-UTR sequence analysis showed that the identified strain belonged to BVDV-la subtype.Analysis of the N gene sequence of BCoV indicated that it was clustered into GⅡb genotype and closely related to the domestic strain.In conclusion,the occurrence of respiratory disease in this farm was mainly caused by M.bovis,accompanied by BCoV and BVDV.Tymycin,lincomycin,and tyvanectin can be preferentially used for prevention and control the disease.The monitoring,prevention and control of viral pathogens should be strengthened.
