摘要

Abstract

Akabane virus(AKAV)is an arbovirus-borne virus,which mainly infetcts ruminants.In recent years,the prevalence of AKAV has become complex in China.To establish a more sensitive detection method for AKAV antibody in a variety of animals,recombination AKAV N protein and HRP labelled recombination AKAV N protein were uesd as the capture antigen and detection antigen,respectively.After the reaction conditions were optimzied,an AKAV antibody double antigen sandwich chemiluminescence enzyme immunoassay detection method was firstly established.The method had no cross-reactivity with the positive serum of BTV,EHDV,BEV and PRV with the intra-and inter-batch coefficients of variation all less than 8%.Compared with the AKAV blocking ELISA kit,the sensibility of the method was increased by 32 folds,with the overall detection time shortened by 42 min,and the κ value of 0.826.Meanwhile,it was proved to be precise by neutralization test.The AKAV antibody chemiluminescence enzyme immunoassay antibody detection method established in this study has strong specificity,high sensitivity,good reproducibility,simplicity,rapidity and precise.This method could be an effective monitoring tool of large-scale serum samples,serosurvey and the prevention of AKAV.
