鸭瘟病毒Cycleave荧光PCR检测方法的建立
Development of a Cycleave Real-time PCR Assay for Detection of Duck Plague Virus
摘要
Abstract
To develop a sensitive,rapid and specific Cycleave real-time PCR method for detection of Duck plague virus(DPV),a pair of specific primers and a Cycling probe were designed according to the DPV UL6 gene sequence deposited in NCBI.The reaction conditions were then optimized for this method.The results showed that the method had good specificity as it did not cross react with other common duck pathogens.The detection limit of this method was 6.5 copies/μL.The repeatability evaluation showed that the coefficients of variation within and between batches were less than 2%.Conclusively,the DPV Cycleave real-time quantitative PCR had high specificity,repeatability and sensitivity,which laid a foundation for the prevention and control of duck plague.关键词
