摘要

Abstract

To investigate the characteristics of Escherichia coli with regional difference,11 E.coli samples were collected from animals in Huairou district and other 22 E.coli samples were collected from animals in Yanqing district for bacterial characterization in the present study.Biochemical reactions,16S rRNA sequencing and MALDI-TOF MS technique were used to identify these E.coli samples.Afterwards,the accuracy of these assays was compared and the peptide mass fingerprint spectrum data were analyzed for these samples.The results showed that the accuracy of three assays was above 95%and the MALDI-TOF MS identification reached more than 99%when multiple sites were tested for one sample.MALDI-TOF MS identification showed good repeatability,high accuracy and fast results especially when the number of test repeats was increased for one sample.Although the E.coli samples from two regions had similar peptide mass fingerprint spectrum characteristics,they were distinguished by clustering and principal component analysis,indicating that the peptide mass spectrums of E.coli samples collected from Huairou and Yanqing had regional differences.This study provided a technical support for traceability analysis of E.coliby MALDI-TOF MS.
