Abstract

Sphingomyelin(SM)and ceramide(Cer)are the main phospholipid components of the Newcastle disease virus(NDV)capsule membrane and sphingomyelin synthase 2(SGMS2)is the regulatory enzyme for SM synthesis.A previous study found significant upregulation of the chicken SGMS2 gene in NDV-infected DF-1,suggesting that the change was associated with NDV infection.In this study,the chicken SGMS2 gene was obtained by cloning and bioinformatic analysis revealed 93.0% and 92.4% homology with human and mouse,respectively.The transmembrane region was highly conserved up to 96.8% across species.The levels of chicken SGMS2 ontology were detected by qPCR in cells and tissues and found to be constant.qPCR showed NDV infection caused dramatic changes in chicken SGMS2 expression levels with significant down-regulation in tissues with high viral content.DF-1 cells were transfected with pCMV6-FLAG-ch-SGMS2 plasmid and significant decreases in mRNA and protein levels of viral NP were found in qPCR and Western blot,indicating that overexpression of SGMS2 inhibited the proliferative effect of NDV.This study laid a solid foundation for revealing the function of chicken SGMS2 protein and also provided a basis for elucidating the role of chicken SGMS2 on NDV proliferation.
