Abstract

In order to understand the prevalence and genetic variation of porcine reproductive and respiratory syndrome virus(PRRSV)in Tianjin,209 tissues were collected from 2021 to 2022 from clinically suspected diseased pigs for detection of PRRSV and ORF5 genetic variation.The results showed that the PRRSV positive rate was 37.3%(78/209).Then,15 representative positive samples from different regions were selected for ORF5 gene cloning and sequencing.Sequence and phylogenetic analysis showed that the 15 PRRSV strains belonged to the American type.The ORF5 genes of 8 PRRSV strains had high homology with the HP-PRRSV-like strains represented by HuN4,and the identities of their nucleotide sequences and deduced amino acid sequences were 89.4%-97.3%and 80.6%-95.5%,respectively while their identities with NADC30,VR2332 and NADC34 were 77.1%-87.1%and 77.1%-86.6%,respectively.The ORF5 gene of 6 PRRSV strains had the highest homology with NADC30-like strains represented by NADC30.The homology of nucleotide sequence and deduced amino acid sequence was 91.9%-92.7% and 91.5%-93.0%,respectively while the homology with HuN4,VR2332,NADC34 and other strains was low at 83.6%-87.8% and 79.6%-86.6%,respectively.The ORF5 gene of one PRRSV strain had the highest homology with NADC34-like strains represented by NADC34.The homology of nucleotide sequence and deduced amino acid sequence of this strain was 96.4% and 95.5%,respectively,while the homology with NADC30,HuN4,VR2332 and other strains was 86.7%-89.9%and 86.6%-92.0%,respectively.The results showed that HP-PRRSV-like and NADC30-like PRRSV were the main epidemic strains from 2021 to 2022 in Tianjin while NADC34-like strains were prevalent,indicating obvious genetic diversity.These results provided a theoretical basis for the prevention and control of PRRS in Tianjin.
