盐酸小檗碱对多重耐药肺炎克雷伯菌抑菌及耐药逆转的体外研究
In Vitro Study on the Bacteriostatic and Drug Resistance Reversal Effects of Berberine Hydrochloride Against Multidrug-Resistant Klebsiella Pneumoniae

Abstract
Objective:To investigate the in vitro bacteriostatic effect and drug resistance reversal effect of berberine hydrochloride against multidrug-resistant Klebsiella pneumoniae(MDR-KP).Methods:Berberine hydrochloride solutions at concentrations of 200,100,50,25,and 12.5 mg/mL were prepared.Then,1 μL of prepared MDR-KP bacterial suspension was added to EP tubes containing different concentrations of berberine hydrochloride solution,and the MIC was determined after incubation.Thirty strains of MDR-KP were induced and cultured in broth containing 50 mg/mL berberine hydrochloride to observe its bacteriostatic effect and evaluate the drug resistance reversal effect on MDR-KP.Antimicrobial susceptibility testing and MALDI-TOF mass spectrometry identification were performed at different time intervals.Results:The minimum inhibitory concentration(MIC)of berberine hydrochloride against 30 strains of MDR-KP was 100 mg/mL.After induction with 50 mg/mL berberine hydrochloride,36.7%of MDR-KP(carbapenem-resistant Enterobacteriaceae,CRE)showed reversed drug resistance,while 63.3%of MDR-KP maintained their resistance profile but exhibited reduced MIC values.MALDI-TOF mass spectrometry analysis revealed that after berberine hydrochloride induction,the peaks at mass-to-charge ratios(m/z)of 2 000 and 7 649 in CRE drug resistance-reversed K.pneumoniae decreased or disappeared,with new base peaks emerging.Conclusion:Berberine hydrochloride exhibits bacteriostatic activity against MDR-KP and can reverse drug resistance in some MDR-KP strains.关键词
