Therapeutic Effect and Mechanism of Schisandrin A on DMH/DSS-Induced Colorectal Cancer in Mice

Abstract
Objective:To investigate the therapeutic effect and mechanism of Schisandrin A on colorectal cancer(CRC)model mice.Methods:Balb/c mice were randomly divided into a normal group,a model group,and Schisandrin A high-dose(160 mg/kg),medium-dose(80 mg/kg),and low-dose(40 mg/kg)groups,as well as a mesalazine group(300 mg/kg).Except for the normal group,mice in the other groups were chemically induced to establish a CRC model using 1,2-dimethylhydrazine(DMH)and dextran sulfate sodium(DSS).The treatment groups were administered different doses of Schisandrin A or mesalazine by gavage,while the normal and model groups received an equal volume of distilled water.The treatment lasted for 10 weeks,with daily administration.The general clinical symptoms and histopathological changes in colorectal tissues were observed.The expression level of proliferating cell protein 67(Ki-67)in mouse colon tissues was detected by immunohistochemistry,and the expression of PI3K and Akt1 in the colon was measured by Western blot.Results:Compared with the normal group,the model group showed increased DAI scores,significantly shortened glass bead expulsion time,and reduced colon length.After Schisandrin A administration,the DAI scores decreased,glass bead expulsion time prolonged,and colon length gradually recovered.Pathological results demonstrated that Schisandrin A significantly improved abnormal hyperplasia of colorectal epithelial cells,glandular structural abnormalities,and inflammatory cell infiltration in CRC mice.Immunohistochemical results showed that Schisandrin A significantly reduced Ki-67 expression in the colon of CRC mice.Western blot results revealed that,compared with the model group,the expression of PI3K and Akt1 in the colon of Schisandrin A-treated mice was significantly decreased(P<0.01).Conclusion:Schisandrin A may inhibit the abnormal proliferation of colorectal cancer cells,alleviate chronic inflammation,and reduce cancer occurrence in CRC mice by suppressing the activation of the PI3K/Akt signaling pathway.关键词
