Identification and Analysis of a Mixed Infection of Six Viruses in Sweet Potato Cultivar"Mashali"

Abstract

Transcriptome sequencing and RT-PCR were performed on four sweet potato samples collected from Linyi City,Shandong Prov-ince,to study the types of viruses that infected sweet potato"Mashali",in order to provid references for the breeding of virus-free sweet potato seedlings and the formulation of prevention and control strategies.The sweet potato samples of"Mashali"showed obvious mosaic and crinkling symptoms.Six known viruses,including sweet potato feathery mottle virus(SPFMV),sweet potato laten virus(SPLV),sweet potato leaf curl virus(SPLCV),sweet potato virus G(SPVG),sweet potato virus C(SPVC)and sweet potato virus 2(SPV2)were identified.The specific primers of the above viruses were selected for RT-PCR detection,the target bands with the expected size of the six viruses were obtained,and the target virus sequences were verified by sequencing.The results of RT-PCR and transcriptome sequencing were consistent with the results of 3 samples containing 6 viruses and 1 sample containing 4 viruses.The phylogenetic analysis based on the CP gene showed that the detected SPFMV had a close evolutional relationship with SPFMV russet crack(RC),the detected SPLCV,SPV2 and SPVC were most closely related to the isolates of China,SPVG was most closely related to the isolates of CH1,and there were significant differences among isolates from SPLV collected from different region.The results of this study will provide theoretical basis for the establishment of field control and virus-free tech-nology system of sweet potato cultivar"Marasali".
