Abstract

This paper explores the extraction and purification process of total saponins from Panax japoni-cus stems and leaves.The contents of total saponins,ginsenoside Rg1,ginsenoside Ⅴ,ginsenoside Ⅳ and ginsenoside Ⅳa in the stem and leaves of Panax japonicus are determined by colorimetric method and high performance liquid chromatography(HPLC);the optimal extraction process of saponins in the stem and leaves of Panax japonicus is derived by using single factor experiment and Box-behnken method;and the purification process of the saponins in the stem and leaves of Panax japonicus is investigated by using a macroporous resin adsorption method.The results show that the total saponin content in the stems and leaves of Panax japonicus is 8.90%by the vanillin-acetic acid spectrophotometry method.The contents of ginsenoside Rg1,ginsenoside Ⅴ,ginsenoside Ⅳ and ginsenoside Ⅳa determined by HPLC are 7.8,20.9,11.5 and 3.0 mg/g respectively,indicating that the stems and leaves of Panax japonicus have certain de-velopment prospects.With temperature,ethanol volume ratio and material-liquid ratio as the dependent variables,the optimal process for extracting the total saponins from the stem and leaves of Panax japonicus by using Box-Bencken method is obtained.That means the ratio for material and liquid is 1∶22(g/mL)with an ethanol volume ratio of 78%and an extraction temperature of 71℃.The optimal adsorbent of D-101 is screened out,and the best purification process is obtained.Adjust the pH value of the aqueous so-lution of crude extract of medicinal herbs to neutral,after the sample loading,elution with water,20%eth-anol and 80%ethanol sequentially,collection of 80%ethanol elution site,recovery of ethanol,and drying under reduced pressure to obtain the total saponins extract from the stem and leaves of Panax japonicus.After purification,the content of total saponin extract from the stem and leaves of Panax japonicus increa-ses from 8.90%to 33.41%,and the total saponin yield is 75.52%.The results show that the stem and leaves of Panax japonicus contain similar active ingredients as those in Panax japonicus roots,and the ex-traction and purification can achieve the effect of enrichment,which provides the basis for the comprehen-sive use of Panax japonicus stems and leaves in basic research.
