摘要

Abstract

[Objective]Proper carrier and activator for formulating a Metarhizium anisopliae Ma54-based biocontrol agent against Ectropis grisescens on tea bushes were investigated.[Method]Suspension of M.anisopliae Ma54 spores was mixed with kaolin,diatomite,rapeseed meal,perlite,or activated clay.Spore viability and germination rate of the mixtures after storage was measured by colony count on PDA plate observed under a microscope.Trehalose or tyrosine at 6 gradients of concentration,i.e.,0.01%,0.10%,0.20%,0.30%,0.40%,and 0.50%,were added to the PDA as an activator to examine the effect in promoting spore germination.The finalized mixture was extruded to form granules with Ma54 spore survival rate determined.[Result]None of the 5 tested carriers exerted significant effect on the spore germination.Nonetheless,perlite and kaolin extended the granule shelf life with a spore germination rate of 61.47%and 62.87%,respectively,after a 28d storage.Those rates were significantly higher than 21.07%of the counterparts without the carriers.The presence of either trehalose or tyrosine in the granules significantly increased the spore germination rate by 20.84%over control.Addition of either kaolin or perlite significantly extended the shelf life of the granules.However,the former caused 70.18%reduction on the spore vitality by the granulation process,whereas the latter induced merely a loss of 21.37%.[Conclusion]Overall,perlite was considered the preferred carrier and tyrosine the choice activator for making Ma54 spore granules as a biocontrol agent.
