Preparation and Properties of Alginate/poly(N-isopropylacrylamide)-Polylysine Composite Hydrogel

Abstract
It is well known that single alginate(Alg)hydrogels suffer from weak mechanical strength,lack of cellular recog-nition sites,excessive swelling and uncontrolled degradation,and thus fail to meet the needs of ideal scaffolds.To improve the performance of alginate hydrogels,homogeneous alginate/poly-N-isopropylacrylamide-polylysine(Alg/PNIPAAm-PLL)composite hydrogels were successfully constructed using a combination of interpenetrating network technology,poly(lysine)(PLL)electrostatic assembly,and hydroxyapatite/D-(+)-glucono-delta-lactone endogenous cross-linking.Mean-while,the effects of the mass ratio of PNIPAAm to Alg on the microscopic morphology,compressive strength,swelling and biocompatibility of the constructed Alg/PNIPAAm-PLL composite hydrogels were investigated.The results showed that the PNIPAAm mass concentration of 1.5%significantly enhanced the mechanical properties of Alg/PNIPAMM-PLL composite hydrogels compared to single Alg gel scaffolds.In addition,the formation of IPN structure limited the excessive swelling of Alg/PNIPAAm-PLL to a certain extent,and the interactions between the components of the composite hydrogel as well as its own temperature sensitivity played a role in the swelling property.The results of the in vitro biological evaluation experi-ments showed that MC3T3-E1 cells were able to adhere,proliferate and differentiate on the Alg/PNIPAAm-PLL composite hydrogel with good biocompatibility,but the excessive amount of PNIPAAm would reduce the cell viability,impede the cell growth,and be detrimental to the transfer of nutrients and metabolic wastes from the cells,thus reducing the cell viability.Therefore,alginate hydrogels constructed by combining the interpenetrating network technology with PLL electrostatic as-sembly have a broad application prospect in biomedical field.关键词
